Principle:
Peptone and beef extract powder provides carbon, nitrogen and vitamins to meet the needs of bacterial growth; sodium chloride maintains osmotic equilibrium; calcium carbonate can neutralize bacterial acid production and absorption of toxic metabolites; bile and brilliant green E. coli and other groups can inhibit Gram-positive bacteria.
Formulation (per litre):
Peptone 9g
Beef extract powder 4.5g
Sodium chloride 2.7g
Calcium carbonate 40.5g
Anhydrous sodium thiosulfate 31.9g
Ox bile 5g
Final pH7.0 ± 0.2
How to use:
- Suspend 93.6g in 1L of distilled or deionized water. stirring heated to boiling until completely dissolved, packed in flasks.
- Place the flask into a sterile pot, autoclave at 121 ? for 20min, cooled to 30 ?, adding a supplement.
- Diluted and treated samples.
Quality control:
Quality control strains were inoculated,and cultured at 36 ± 1
oC for 24h , results show as follows:
