For rapid isolation and identification of Salmonella.
Principle:
Ability antibacterial agents against Gram-positive bacteria, and enhance the medium inhibit bacteria; peptone and yeast extract powder provides carbon and nitrogen sources and trace elements; sodium chloride maintains osmotic equilibrium; agar as culture medium coagulant; mixing pigment with Salmonella and coliforms, respectively corresponding to the occurrence of a specific enzyme reaction, hydrolysis of the substrate, the release of the color groups in the pale yellow tablet Salmonella produce magenta colonies, coliform produce blue-green colonies.

How to use:
- Suspend 47.5g the product, dissolved in 1000ml of distilled water, mixed and heated to 100 ?, stirring constantly until the regular press completely dissolved (can not be heated to high temperatures sustainably, it is recommended not to repeat the boiling solvent), without autoclaving, cooled to 50 ?, pour on sterilized dishes.
- Prepare fluid sample and enrichment broth according to the national standard, SN standards, FDA standards or other sample preparation methods.
- A two-step enrichment method was recommended:
- Pre-enrichment: place 25g of the treated sample in 225mL buffered peptone water and mixed uniformly at 37 ? cultivating for 6 ~ 8h;
- Selective enrichment: Take 1mL of Pre-enrichment broth and adding to 9mL of four brilliant green broth, sodium-sulfur enrichment broth (TTB), and after mixing 37 ? cultivating for 18 ~ 24h;
- Take one loop of the enrichment broth with inoculation loop, streaked onto Salmonella colour tablet, placed in 36 ± 1 ? cultured for 18-24 hours.
- Observe the results. picked magenta colour, flat and transparent, smooth edges, about 2mm in diameter suspicious colonies for further testing on the plate.
- MicrogenTMGN-ID or API20E or biochemical test tube was used for suspected biochemical identification of Salmonella colonies.
Quality control:
This product appears light yellow after the pour on a plate, these strains were inoculated after 36 ± 1 ? cultured 18-24h growth in the following table:
