Bacillus Cereus Chromogenic Medium (Chromogenic Bacillus Cereus Agar)

Usages:
For rapid isolation and identification of Bacillus cereus. in a food sample.
 
Principle:
Peptone and yeast extract powder provides carbon and nitrogen sources and traces elements; disodium hydrogen phosphate and potassium dihydrogen phosphate as a buffer; agar as medium coagulant; mixing pigment with Bacillus cereus enzyme-specific reaction, hydrolysis of the substrate, the release of the colour groups, in a pale yellow wax-like tablet produced by Bacillus green - blue-green colonies.

Peptone:15g
Yeast extract powder:4g
Disodium hydrogen phosphate:2.52g
Potassium dihydrogen phosphate: 0.28g
Agar:15g
Mixing pigment: 3.25 g
Final pH 7.2 ± 0.2
 
How to use:

1. Suspend 40 g of the product, dissolved in 1000ml of distilled water, heated to boiling with stirring until completely dissolved, dispensing into the flask, autoclave at 121^oC for 15min, cooled to 50^oC, additives (SR0210) were added with 1ml of sterile water, dissolved completely, followed by addition of 100 ml medium, mix, pour sterilized dishes.
2. With sterile saline or phosphate buffer preparation sample dilutions.
3. Take 0.1ml of each dilution, inoculate Bacillus cereus on agar plates coated with colour or crossed.
4. After culturing at 37^oC for 24 hours, select the appropriate plate number of colonies and counted.
5. Validation tests for further confirmation of the presumptive Bacillus cereus, such as oxidase, Gram staining.
6. Report number (ml) of food samples: Bacillus cereus per gram.

 
Quality control:
This product appears light yellow after pour on plate, these strains were inoculated after 36 ± 1^oC 18 ~ 24h culture growth in the following table.
